Summary of Microarray Experiments

Experiments were performed on rat U34A Affymetrix chips and on human HG-U133A chips in collaboration with the group of T. Orntoft (Molecular Diagnostic Laboratory, Aarhus University Hospital, Aarhus, Denmark). Experiments performed on rat U34A Affymetrix chips: Time course experiment for cytokine-induced genes in INS-1E cells were exposed for six different time points: 1, 2, 4, 8, 12 or 24 hours to IL-1β (10 U/ml) + IFNγ(100 U/ml) +/- the inducible NO synthase (iNOS) blocker N(G)-monomethyl-L-arginine (NMA) NMA (1.0 mmol/l). Results are means of 2 experiments for each time point. 698 cytokine-responsive genes will be included by February 2005 in T1DBase from this experiment.
Cytokine-induced genes in primary rat beta-cells: FACS-purified rat beta cells were exposed for 6 hours or 24 hours to IL-1beta (50U/ml) or a combination of IL-1beta (50U/ml) and IFN-gamma (1000U/ml). Results are fold change compared to controls and represent means of 2 chip determinations of one pool from 4-6 independent experiments or in the case of IL-1beta and IFN-gamma, mean determination from 2 pools from 4-6 independent experiments, each analyzed in duplicate.
Double stranded RNA- and IFN-gamma-induced genes in primary rat beta cells: FACS-purified rat beta cells were exposed for 6 hours or 24 hours to IFN-γ (1000U/ml) and/or Poly-I:C (polyinosinic-polycytidylic acid; 100 µg/ml). Results are fold change compared to controls and represent means of 2 chip determinations of one pool from 4-6 independent experiments.
Experiments performed on human U133 Affymetrix chips: Global profiling of coxsackievirus- and cytokine-induced gene expression in human pancreatic islets: Isolated human islets were exposed for 48h to either IL-1β (50 U/ml) and IFN-γ (1000 U/ml) or CBV-5 (coxsackievirus 5B; multiplicity of infection 30-100). Results are fold change compared to controls and represent means of 3 chip determinations from 3 individual donor samples.
